Abstract This study was performed in the laboratory of Intra Cytoplasmic Sperm Injection and freezing of sperm in the fertility center of Al-Sader Medical City / Al-najaf Alashraf City during the period from May, 2012 to April 2013. The study aimed to study the effect of rapid freezing and thawing processes on semen and sperm parameters, Malondialdehyde concentration ( MDA) and abnormal sperm chromatin percent, besides to study the effect of adding types from antioxidants Including vitamin E in concentrate 40 and Glutathione in concentrate 1 mm to the sperm freezing media (Sperm Freeze) In minimizing the negative effects which result from sperm rapid freezing and thawing processes.
The study included three types of Infertility patients Normozoospermic, Oligozoospermic, and Asthenozoospermic patients. The Number of patient’s was (30) in each case . . The Mean of age of Normozoospermic patients were (32.2+_1.31) year, Oligozoospermic patients (31.1+_1.29) year and, Asthenozoospermic patients (33.1+_1.23) year. . The results showed that the sperm activation processes lead to a significant difference (p<0.05) in sperm parameters in comparison with these before activation . The present study results showed that sperm activation by using simple layer technique to patients samples of Normozoospermic and Oligozoospermic patients lead to a significant decrease (p<0.05) in sperm concentration, round cell concentration , Malondialdehyde concentration and abnormal sperm chromatin percent Means and a significant increase (p<0.05) in sperm motility forward movement percent , Sperm viability percent and normal sperm morphology percent in comparison with their values before activation . While the sperm activation processes of Asthenozoospermic patients by useing Centrifugation Wash – Out technique lead to a significant decrease (p<0.05) in sperm concentration, round cell concentration and abnormal sperm chromatin percent Means and a significant increase (p<0.05) in sperm motility forward movement percent, sperm viability percent, normal sperm morphology percent and Malondialdehyde concentration In comparison with their values before activation. . The present study results showed that sperm rapid freezing processes of patients who were included in the study by using sperm freezing media ( Sperm Freeze) and thawing processes after one month from freezing leads to a significant decrease (p<0.05) In the sperm concentration, sperm motility forward movement percent, sperm viability percent and normal sperm morphology percent. and a significant increase (p<0.05) in the Malondialdehyde concentration and abnormal sperm chromatin percent and no significant difference(p<0.05) in round cell concentration in comparison with their values after activation .
Study of freezing time by calculating sperm parameters in the first month and compared it with second and third months after freezing and thawing in the samples of patients who were included in the study showed there was a there was a significant decrease (p<0.05) in the sperm motility forward movement percent and sperm viability percent ,and there was a significant increase (p<0.05) in the Malondialdehyde concentration and abnormal sperm chromatin percent and no significant difference(p<0.05) in the sperm concentration, normal sperm morphology percent and round cell concentration Means in the second and third month In comparison with their values after doing the thawing processes after one month from freezing . The study results shows a significant decrease (p<0.05) in the sperm motility forward movement percent and sperm viability percent ,and there was a significant increase (p<0.05) in the Malondialdehyde concentration and abnormal sperm chromatin percent and no significant difference(p<0.05) in the sperm concentration, normal sperm morphology percent and round cell concentration when comparing third month Means with second month Means. . The study results shows that used of sperm Freezing Media ( Sperm Freeze) with adding antioxidants like vitamin E or Glutathione in single form or in the mixed form from the tow antioxidants leads to a significant difference (p<0.05) in sperm parameters of study included patients , there was a significant increase (p<0.05) in the sperm motility forward movement percent s and sperm viability percent ,and there was a significant decrease (p<0.05) In The Malondialdehyde concentration and abnormal sperm chromatin percent and no significant difference (p<0.05) In the sperm concentration, normal sperm morphology percent and round cell concentration In comparison with their Means when used of sperm freezing media ( Sperm Freeze) alone . . It can be concluded from this study that sperm rapid freezing and thawing processes had a negative effect on the semen and sperm parameters and Malondialdehyde concentration and abnormal sperm chromatin percent in all samples of study included patients , and this negative effect of rapid freezing and thawing processes increased with increasing of freezing time , and Adding of antioxidants vitamin E or Glutathione in single form or In the mixed form shows resistance to this negative effect leads to a significant increase (p<0.05) In the studying parameters .
